Validation of cell-surface SLC35D3 expression in cancer cell lines by flow cytometry and comparison with CCLE transcriptomic data. Flow cytometry histograms of cell-surface SLC35D3 staining in human cancer cell lines (HCT 116, LoVo, QGP-1, NCI–H1930, and SNU-16). For each cell line, the MFI ratio (anti-SLC35D3/isotype) and the corresponding mRNA expression level (log2[TPM+1]) from the CCLE are indicated below the histogram. HCT 116 served as a negative control and showed minimal surface staining, consistent with a CCLE value of log2[TPM+1] = 0.0.

Synergistic combination of EdU + CF10 causes mitotic catastrophe in HCT116 cells. ( A ) Dot plot representations of flow cytometric analyses of pH3 levels with the indicated treatments for 48 h. Total DNA content was detected with propidium iodide ( x -axis) with anti-pH3 staining ( y -axis). ( B ) Quantification of phospho H3 + cells for treatments in panel (A). Single-agent EdU showed increased phospho H3 + cell population only in G2/M cells, whereas single-agent CF10, SD-1, and SD-2 showed significant phospho H3 positive cells in both G1 and S phase, consistent with premature chromatin condensation. ( C ) Confocal micrographs of representative spindle fiber assembly images from each treatment group with microtubules in spindle fibers stained with antibody specific for α-tubulin, conjugated with Alexa-647 fluorophore (red). The pericentromeric regions of the chromosomes were stained with antibody targeting pH3, conjugated with Alexa-488 (green). Nuclei were stained with DAPI (blue). A monopolar spindle fiber is shown from a representative SD-1 cell, while representative images from SD-2 treated cells highlight a multipolar spindle fiber assembly. Only in cells treated with SD-1 or SD-2 was abnormal spindle fiber formation detected confirming mitotic catastrophe. Magnification was 100× with oil immersion; scale bar is 10 μm.
